After sacrifice with an overdose of anesthetic (12 ml/kg 20% urethane; Hengyuan Biological Technology Co., Ltd., Shanghai, China), the cartilage was sliced into small pieces from the femoral trochlea and digested with 2.0 mg/ml type II collagenase (Invitrogen; Thermo Fisher Scientific, Inc., Waltham, MA, USA) at 37?C for 4 h. Then, the digest solution was filtered with 200 mesh?filtrating screen to remove large fragments.
